A 19-year-old man has died after the vehicle he was driving overturned after hitting an animal on the Buntine Highway overnight.

Police at Kalkaringi received a report of the crash 120km east from Kalkaringi on the Buntine highway at 11pm.

At this stage it appears the interstate driver has hit a kangaroo causing his Toyota Hilux to leave the carriage way and roll numerous times off the side of the road.

A 19-year-old female passenger received minor injuries from the crash and is currently assisting police with further information.

Katherine crash investigators are travelling to the scene and will examine if speed or alcohol were contributing factors and if seatbelts were worn.

Police and Northern Territory Emergency Services members are at the crime scene, which is being maintained throughout the day.

Motorists using the Buntine Highway, in particular heavy vehicles, are urged to exercise caution while the crime scene in established.

Territory Duty Superintendent Michael Murphy said drivers not familiar with Territory roads and conditions need to take extra caution to avoid such tragedies.

"It's unfortunate it takes a death such as this to remind drivers to take care on our roads, said Superintendent Murphy

"Both visitors and locals need to remember Territory roads present long distances between destinations, different road surfaces and wildlife on roads.

"Police urge motorists to plan their trip for daylight hours rather than travel in lowlight conditions."

In a report released in 2006, visitors accounted for 20 per cent of fatalities and 19 per cent of trauma in the NT from 2000 - 2005.

"This is a timely reminder for motorists to adhere to speed limits and rest when fatigued."

A brief will be prepared for the Northern Territory coroner.

Both families have been advised of the tragedy.
